ROLE OF THE COMPLEMENT PATHWAY IN SKIN DISEASES

Abstract


The complement system is a collection of circulating, membrane-bound proteins that play an important role in defense against microbes. Complement proteins are proteolytic enzymes and complement activation involves sequential activation of these enzymes. The complement pathway can be activated by one of 3 pathways, namely the alternative pathway, the classical pathway, and the leptin pathway. Complement is involved in several skin diseases such as psoriasis, acne vulgaris, hidradentitis suppurativa, SLE, urticaria, urticaria vasculitis, and bullous pemphigoid. Several causes including auto-antibodies and micro-organisms can activate complement, either through classical or alternative pathways. The main objective of this review is to provide an overview of the role of complement in inflammatory skin diseases and to discuss the rationale and potential targets for pharmacological intervention using complement inhibitor therapy.
